I played Sentient Ink Pot when I had already played Nameless Nightmare, and had not filled any spots on it yet. The ink spot on Nameless Nightmare did not fill. On my next turn, I gained ink, and was not given the option to fill the ink spot on Nameless Nightmare.

This is true, but you hadn't filled the prerequisite spot (the red) on Nameless Nightmare.

True, as described in the rules. You need to fill the prerequisite spots and then the ink will fill in.
